    Towards A Better Understanding of Orofacial Pain Pathology and Treatments

    Presentation Description

    Migraine and temporomandibular joint disorder (TMD) are prevalent, debilitating orofacial pain conditions that are often comorbid. Migraine and TMD pathology involves activation of trigeminal ganglion neurons, enhanced neuron-glia communication, and development of peripheral and central sensitization. In my presentation, I will describe our preclinical models of chronic migraine and TMD that involves trigeminal sensitization caused by reported risk factors such as neck muscle inflammation and sleep deprivation. In this sensitized state, trigeminal nociception can be triggered by a pungent odor (migraine) or prolonged jaw opening (TMD). I will discuss our recent findings that daily supplementation by the nutraceuticals, grape seed extract and chicken bone broth, or non-invasive vagus nerve stimulation (nVNS) inhibit pain signaling in our preclinical models of migraine and TMD. Dietary grape seed extract and nVNS mediate cellular and molecular changes in the spinal cord to enhance the GABAergic and serotonergic descending inhibitory pain pathway. Dietary inclusion of the chicken bone broth increases the level of the commensal gut bacteria Bacteroides acidifaciens, which produces the anti-inflammatory short chain fatty acid acetate. In addition, I will share recent results that the burdensome symptoms of hearing loss and hyperacusis are observed in our preclinical models.
